Ebola virus disease (EVD) is an endemic disease caused by Ebola virus (EBOV) with a high fatality rate and long-term sequelae that would require a systematic chronic treatment. All the possible EBOV targets have to be explored, in the need of new versatile and cost-effective anti-EBOV therapeutics. The IFN-beta pathway is critical for counteracting viral infections and, EVD fatal outcome is strongly related to the EBOV ability to hijack the innate immune response. EBOV VP35 and VP24 are key factors for viral replication and IFN-beta suppression, and also validated drug targets. Several small molecules were identified as able to block these viral components involved in immune evasion, inhibiting also viral replication, showing that an antiviral approach targeted at the restoration of the innate immune response is a feasible and promising goal.
